MazF-ssrA
The MazF endotoxin, codon optimized, with a native ssrA tag (ANDENYALAA) appened on the C-terminus.
The MazF endotoxin is an endoribonuclease that will cleave mRNAs at ACA triplets. Cleavage of the mRNAs will prevent proper translation, resulting a increased mRNA degradation, impaired tranlsation, and ribosome stalling upon non-stop mRNAs. Expression of this toxin, particularly at levels beyond that of the cognate anti-toxin MazE, will thus lead to arrest of cell growth and eventually cell death.
The ssrA tag was added to promote rapid degradation of MazF, hopefully preventing any leaky expression of MazF from being toxic.
